Legacy Analytics Applied to Emerging Therapeutic Modalities

Many analytical technologies previously considered groundbreaking have become commonplace in the modern laboratory supporting therapeutic discovery, development, and manufacture. No better example of this is in the field of mass spectrometry, which has evolved from the exclusive research domain of the organic chemist to a tool that is almost ubiquitous in the testing of many therapeutic types. Indeed, most current MS systems have developed largely as a result of demands from the bio/pharmaceutical industry. This has led to innovative designs for sample introductions such as Desorption Electrospray Ionization (DESI), and Laser Ablation. Novel ion separation mechanisms such as ion-mobility have also evolved to expand the capabilities of mass spectrometry.

Other well-established techniques, specifically those used in the evaluation of higher order structure, have also come under increased attention with a focus on improvements in performance such as sensitivity and robustness. Equipment manufacturers have responded by developing instruments such as the Microfluidic Modulation Spectrometer which provide greater sensitivity and throughput but nevertheless remains reliant of long-established IR-based technology. 

The almost constant advancement of emerging drug modalities results in ever increasing challenges to analytical technologies, many of which are successfully addressed by adapting and modernizing legacy technologies. High resolution mass spectrometry, a technique born in the last century to provide proof of structure for small molecules, has now become widespread for accurate mass measurement of high mass biopolymers and resolution of complex mixtures.

This webinar provides a review of some of these methodologies and describes how improvements are allowing sustained support to the contemporary drug industry.

Christopher Ziegenfuss
BioPharmaSpec Inc

Chris Ziegenfuss is the Vice President at BioPharmaSpec Inc. and has 25 years of experience working in the CRO field, primarily in the application of mass spectrometry and higher order structural characterization techniques to the analysis of a wide variety of pharmaceutical and biopharmaceutical products. Formerly, a member of the M-Scan group, he played a key role in establishing BioPharmaSpec’s US laboratory in 2015. In his current role, Ziegenfuss oversees BioPharmaSpec Inc’s laboratory operations, including the design and management of structural and physicochemical characterization studies for both novel and biosimilar biopharmaceuticals, including assessments of amino acid sequence, post-translational modifications (PTMs) such as glycosylation, higher order structure analysis and aggregation analysis.
