The advent of ultra-high-pressure LC (UHPLC) and its successful commercialization in the last few years has brought forth a modern HPLC platform capable of higher speed, resolution, precision and sensitivity.  Currently, all major HPLC manufacturers have some type of low-dispersion UHPLC products with upper pressure limits ranging from 10,000 to 19,000 psi. 

This paper describes a number of popular myths or half-truths in ultra-high-pressure liquid chromatography (UHPLC) and provides data or case studies that delineate, contradict or even repudiate some of these popular beliefs.  These are:

  1. Viscous heating is a huge issue for sub-2um particle columns of inner diameters > 2 mm.
  2. Binary high-pressure mixing pump is a “must” for a low-dwell volume system.
  3. One can reduce dwell volume to near zero without performance issues.
  4. UHPLC provides substantially higher sensitivity than HPLC.
  5. Method transfer from UHPLC to HPLC is “a piece of cake” and method re-validation is not needed.
  6. One needs gold plated fittings with double ferrules in UHPLC to withstand the pressure.
