• The most important and primordial role of proteins in living systems is to perform energy transduction.
• The roles of protein as energy transduction machines is enabled by ionizable residues buried in hydrophobic environments.
• The properties of buried ionizable groups are highly anomalous and consequences of ionization of these buried groups are complex.
